1,3-Butanediol as a therapeutic for kidney disease

Background

Approximately 33 million Americans have kidney disease and over the last decade, the rate of chronic kidney disease has risen more than 25%. Kidney disease can cause additional complications including high blood pressure. For example, 80%-90% of CKD patients have high blood pressure at some point during their time with the disease and are often prescribed ACE inhibitors and ARBs.

 

Invention Description

It is suggested in the literature that the Nlrp3 inflammasome is a possible therapeutic target in the treatment of progressive chronic kidney disease. Researchers found that 1,3-Butanediol protects the kidneys through mediated inhibition of Nlrp3-inflammasome activation. It acts to lower hypertension by increasing Betahydroxybutyrate (BHB), thereby blocking the Nlrp3 inflammasome. Treated animals had decreased kidney fibrosis and improved kidney function. In vivo studies demonstrated lower levels of renal Nlrp3, Casp1,  IL1b, IL18, Lcn2, etc. in high-salt diet rats treated with 1,3-butanediol. Protein casts and macrophage infiltration were also significantly lower in kidneys of treated animals.
